The objective of plant breeding is to develop stable plant varieties which are high in productivity and resistant to abiotic ( salt, drought, and heat) and biotic (fungal and bacterial) stress factors. Laborous and time consuming breeding programmes are continous debate due to changes in ecological conditions and market demands. Classical breeding relays on combinations, recombinations and selections of plants. Selection process usually depends on phenotypic characteristics and thus highly affected from changes in environmental conditions that have adverse effect on the success of breeding programme.
The interest of our group is to isolate and characterize rust disease (Puccinia spp.) resistance markers, the disease is known to be the biggest problem that cause drastic loss in the yield of wheat production, especially in Central Anatolian Region of Turkey. For this purpose, we have used rust disease resistant isogenic lines of wheat which were developed by introgression of resistance trait from Aegilops species and we have performed polymorphism analysis using RAPD-PCR approach. Two separate resistance markers namely sspJ14 ans sspA15 were isolated, cloned and sequenced. These marker DNAs are going to be good candidates for marker-assisted selection (MAS) during breeding programmes. Currently we are working to show their close linkage to resistance trait by finding the localization of markers on introgressed chromosome.
The other interest of our group is to establish DNA-based test service to provide DNA-fingerprinting, phylogenic relation and disease-resistance analysis for commercially valuable species (such as sunflower and cherry). Currently we are optimizing the application of PCR-based methods using RAPD, microsatellite, chloroplast-specific SCARs and Orobanche disease resistance -specific primers. Availability of such service will provide an opportunity for those govermental and private organizations to improve their breeding programmes with the knowledge provided by such modern molecular techniques and to develop high quality export products that can be in demand by foreign markets.
